The plasmid complement of Lactobacillus helveticus ATCC15009 (plasmids pLH1, pLH2 and pLH3) has been sequenced and is being analysed for functional properties. Sequences used include Accession Numbers X62150 (1-427), X72021 (21-2404 and 6891-10334), X17655 (141-2404), U83488 (260-6674), U00453 (2394-9915), L07895 (10544-12904) modified in the present laboratory. Additional sequences generated were deposited under Accession Numbers AF301605 9714-10561 and from 12905 to 14284 (unpublished). The remaining sequence was taken from Accession Number L39769 (14285-22421). All three plasmids share common sequences. Intact or degenerate IS elements are present on two of the plasmids only. It has been noted that the smallest plasmid probably encodes an element from the IS982 family of mobile genetic elements and multiple copies of this were present on the chromosome of all nine strains of Lb. helveticus tested. The use of strain ATCC15009 as a host for mobilizable recombinant plasmids derived from conjugative plasmid pIP501 is also reviewed in relation to the exploitation of this strategy to introduce new genetic material in order to study gene expression in this industrially important species.
